A groundbreaking study published in January 2026 has identified a clear genetic signature in "super agers": people over 80 with memories as sharp as those decades younger. The research reveals they possess a double genetic advantage: a lower likelihood of having the chief Alzheimer's risk gene and a higher prevalence of a protective variant.
The study, the largest of its kind, analyzed data from 18,080 individuals. Researchers found super agers were 68% less likely to carry the APOE-ε4 gene variant, the strongest genetic risk factor for late-onset Alzheimer's, compared to octogenarians with Alzheimer's dementia. Strikingly, they were also 19% less likely to carry APOE-ε4 than cognitively healthy peers their own age.
Conversely, super agers were 28% more likely to carry the protective APOE-ε2 variant compared to typical older adults and over 100% more likely than those with Alzheimer's. "This suggests the super-ager phenotype identifies a particularly exceptional group with reduced genetic risk," said lead author Dr. Leslie Gaynor of Vanderbilt University Medical Center.
The findings, drawn from eight national aging cohorts, provide robust evidence that genetics contribute to remarkable cognitive resilience. Scientists hope studying these genetic advantages will unlock new pathways for preventing or delaying Alzheimer's disease.
